The synthetic tetrapeptide YVAD (Tyr-Val-Ala-Asp) conjugated to p-nitroanilide (YVAD-pNA) is a colorimetric substrate for caspase-1 enzyme activity. YVAD-AFC (YVAD conjugated to 7-amino-4-trifluoromethyl coumarin) and YVAD-AMC (YVAD conjugated to Amino-4-methyl coumarin) are fluorogenic substrates for the caspase which is detectable following cleavage of the substituted coumarin moiety from the peptide.
The tetrapeptide coupled to fluoromethylketone (YVAD-fmk) is a synthetic inhibitor of caspases. The aldehyde derivative (YVAD-CHO) is another inhibitor.
