This peptide with the sequence EAEDLQVGQVELGGGPGAGSLQPLALEGSLQ is encoded by the human proinsulin gene (Steiner et al, 1967), where it is located between the two chains that subsequently form mature insulin. C-peptide plays a role in the proper folding of insulin, is cleaved from the propeptide to release mature insulin, and is secreted in equimolar amounts with insulin by pancreatic beta-cells, circulating in nanomolar concentrations (typically 0.6 to 3 nM) in healthy individuals. Higher levels are observed in overweight individuals (Polonsky et al, 1988).
