The cDNA for this nuclease was isolated from a human KT-3 leukemic cell cDNA library. CAD mRNA is expressed in a limited number of human tissues, including pancreas, spleen, prostate, and ovary. The CAD gene maps to human chromosome 1p36.3. The enzyme (338 amino acids) cleaves chromosomal DNA during cell death by apoptosis. Overexpression of the enzyme potentiates DNA fragmentation by apoptotic stimuli in several human cell lines.
