This antigen is expressed on pre-B-cells, eosinophils, basophils, and platelets. CD9 is expressed on the plasma membrane of enterocytes (Sincock et al, 1997). The rat antigen (NCA) has been shown to be expressed by astrocytes, oligodendrocytes, ependymal cells, and microglial cells, but not by neuronal progenitor cells and neurons, Kaprielian et al (1995) have demonstrated expression of rat CD9, identified by monoclonal antibodies ROCA1, ROCA2, and B2C11, on peripheral neurons, Schwann cells, and adrenal chromaffin cells.
CD9 plays an essential role in sperm-egg fusion during fertilization (Miyado et al, 2000; Le Naour et al, 2000; Kaji et al, 2000). It is a coreceptor involved in the upregulation of mitogenic and diphtheria toxin binding activities of membrane-bound HB-EGF (Cha et al, 2000; Nakamura et al, 2000). CD9 also acts as a suppressor of metastasis. Melanoma cells engineered to express CD9 show a reduced metastatic potential (Ikeyama et al, 1993). High levels of CD9 expression in the primary tumor correlates with the appearance of metastases in melanomas, colon, lung, and breast cancers (Si and Hersey, 1993; Miyake et al, 1995; Adachi et al, 1998; Mori et al, 1998).
