This immediate-early gene product encoded in the genome of human cytomegalovirus (Tennet and Colberg-Poley, 1991) suppresses antigen presentation to CD8(+) cytotoxic T-lymphocytes and therefore acts as an immunoevasin, hindering viral clearance by cytotoxic T-cells. US3 binds and retains newly synthesized MHC-1 proteins in the endoplasmic reticulum and blocks tapasin-dependent peptide loading (Jones et al, 1996; Park et al, 2004). Noriega et al (2012) have reported that US3 modulates destruction of MHC-1 molecules in cooperation with another virus-encoded protein, Cytomegalovirus US11 protein.
