this assay has been designed to allow quantification of cell death by apoptosis. It detects diffusion of small DNA fragments from apoptotic cells. These fragments are generated under alkaline conditions from apoptotic cell DNA, which conain abundant alkali-labile sites. The diffused DNA fragments are detected by a fluoresent dye (e. g., YOYO-1). Apoptotic cells show a circular gradient of granular DNA with a dense central zone and a lighter and hazy outer zone, giving the overall appearance of a halo.
