ESMD is a short tetrapeptide sequence (amino acids 25-29) in the inactive 32 kDa precursor of the apoptotic cysteine protease, caspase-3. Cleavage after this site, together with cleavage at another site, yields the two subunits of bioactive caspase-3 during cell death by apoptosis (Nicholson et al, 1995). Ac-ESMD-CHO (acetyl-ESMD-aldehyde) can be used as a potent inhibitor of caspase-3 (Han et al, 1997).
General note on chemical modifications: Practically all of the available caspase inhibitors, be they specific or pan-caspase inhibitors, come in chemically modified form. The introduction of an aldehyde group at the C-terminus of the short peptide sequences
