This sequence motif, KENxxxN/D, is a degradation signal (degron) found in many proteins that are targeted for degradation by the anaphase-promoting complex or cyclosome (APC/C), a large E3 RING-cullin ubiquitin ligase assembled from 13 different proteins that controls sister chromatid segregation and the exit from mitosis by catalysing the ubiquitylation of these target proteins, which are then marked for degradation through the ubiquitin proteasome system. The KEN-box motif often occurs together with another degron, D-box. Mutations in the KEN-box motif render proteins resistant to degradation (Pfleger and Kirschner, 2000; for general overview see also: Barford, 201).
