[negative factor] This myristylated protein of 27 kDa is encoded by an open reading frame at the 3' end of the genome of human immunodeficiency virus type 1 (HIV1). The gene is found also in other primate lentiviruses. Nef is a component of the viral particle and is one of the first proteins detected after invasion of the host cell. Nef influences viral infectivity and interactions with host cells in multiple ways (Fackler and Baur; Arora et al, 2002).
Nef has been shown to downregulate expression of CD4, a protein involved in cell activation of T-cells and functioning as a receptor for HIV1 (Greenway et al, 1994). Nef protein also downregulates expression of MHC on the infected cell surface.
