Pep-1 is a designer cell-penetrating peptide (KETWWETWWTEWSQPKKKRKV) (Morris et al, 2001). The KETWWETWWTEW moiety efficiently targets to the cell membrane and forms hydrophobic interactions with proteins. The KKKRKV moiety is derived from the nuclear localization sequence of SV40 large T antigen and improves intracellular delivery and solubility of the peptide vector. SQP is a spacer. The peptide is stable in physiological buffer, lacks toxicity, and is insensitive to serum. Pep-1 is able to efficiently deliver a variety of peptides and proteins into several cell lines (human HS-68 fibroblasts, murine NIH 3T3 fibroblasts
