An assay for IFN-gamma, which measures the concentration of nitric oxide (determined by nitrite accumulation in the culture medium) generated by RAW264.7 cells stimulated with IFN-gamma has been developed. Following the incubation of cells with murine IFN-gamma, NO production is found to be linear at IFN-gamma concentrations between 0 and 10 U/mL and logarithmically linear between 2 and 100 U/mL (Kim and Son, 1996).
