Thymidylate synthetase is a dimeric protein composed of identical subunits. The protein is a critical enzyme for maintaining the dTMP (thymidine-5'-monophosphate) pool required for DNA replication and repair. Thymidylate synthetase catalyzes the reductive methylation of dUMP to generate dTMP, using 5,10-methylenetetrahydrofolate as a cofactor (hence the designation 5,10-methylenetetrahydrofolate:dUMP C-methyltransferase).
