[endoplasmic reticulum aminopeptidase associated with antigen processing] The enzyme is involved in the cleavage of peptides that are displayed as antigenic peptides together with different MHC class I molecules on the surface of potential target cells recognized by killer T-cells carrying the CD8 antigen. ERAAP has a broad substrate specificity, and its expression is strongly upregulated by IFN-gamma. Reducing the expression of ERAAP prevents the trimming of peptides for MHC class I molecules in the ER and greatly reduces the expression of MHC class I molecules on the cell surface. ERAAP thus links initial cytosolic processing of proteins and the generation of final peptides
