K-7/D-6 carries amino acid substitutions in two distinct regions of wild-type IL6 that participate in the interaction with IL6R and shows a 70-fold increased binding affinity as compared to the wild-type molecule (Cabibbo et al, 1994; Toniatti et al, 1996). The protein has an increased biological activityin vivo and in vitro compared to the wild type molecule and has been shown to allow fast hematopoietic reconstitution (significant recovery of thymus, spleen and blood cells) after lethal irradiation and bone marrow transplantation in mice (Frasca et al, 2000).
