This protein of 48 amino acids is found in human and bovine spleen and lymph nodes but not other tissues. It was described originally as Thymopoietin-3. Human splenin differs from human thymopoietin, one of the thymic hormones, at four amino acid positions. A synthetic pentapeptide corresponding to residues 32-36 (Arg-Lys-Glu-Val-Tyr; RKEVY), called Splenopentin (SP5), has the same biological activities as splenin.
