[pancreatitis-associated protein-2] This rat protein is related to PAP-1 (Frigerio et al, 1993). PAP-2 is a member of a multigene family of proteins expressed in several tissues, which includes PAP-1, PAP-3, and lithhostatine. Bodeker et al (1999) have reported that PAP-1, PAP-2, and PAP-3 can form heterodimers.
Alternative splicing of PAP-2 yields two different proteins (Vasseur et al, 1995). The gene is overexpressed by the pancreas during acute pancreatitis but not in intestinal tissues (unlike the related PAP-1 and PAP-3). An elongated form of the protein arising from alternative use of a polyadenylation site, termed C18-2, has been shown to be overexpressed in the pancreas during the development of severe acute pancreatitis in the rat (Honda et al, 2002).
Unlike the related proteins PAP-1 and PAP-3, PAP-2 does not form insoluble thread-like protein aggregates upon cleavage with trypsin. The physiological relevance of this feature remains to be established (Graf et al, 2001).
Zhang et al (2004) have reported that administration of antisense oligonucleotides against PAP-1, PAP-2, and PAP-3 by direct intrapancreatic injection significantly exacerbates the severity of acute pancreatitis in rats and worsens edema, elevates leukocyte infiltration, and induces fat necrosis, suggesting that these proteins mediate significant protection against pancreatic injury.
