This protein of approximately 68 kDa has been described by Doherty et al (1999). Herstatin is derived from an alternatively spliced HER2 (neu, erbB2) transcript that comprises 340 residues identical to subdomains I and II from the extracellular domain of HER2 followed by a unique C-terminal sequence of 79 amino acids encoded by intron 8.
Herstatin binds to the HER2 tyrosine kinase receptor. Herstatin mRNA is expressed in normal human fetal kidney and liver. Reduced levels relative to HER2 mRNA are found in carcinoma cells that contain an amplified HER2 gene. Herstatin appears to be an inhibitor of HER2
