MTT assay

This test is a quantitative colorimetric method to determine cell proliferation. It utilizes the yellow tetrazolium salt [3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium-bromide] which is metabolized by mitochondrial succinic dehydrogenase activity of proliferating cells to yield a purple formazan reaction product. MTS (3-[4,5,dimethylthiazol-2-yl]-5-[3-carboxymethoxy-phenyl]-2-[4- sulfophenyl]-2H-tetrazolium) has been used also in proliferation and cytotoxicity assays, forms an easily detectable formazan product, and has been reported to be more stable than XTT (Goodwin et al, 1995).
