Saporin

abbr. SAP. A toxic protein isolated from Saponaria officinalis. Saporin inhibits protein biosynthesis by binding to a ribosomal protein. Saporin functions as an RNA-N-glycosidase specifically depurinising the 28S RNA of ribosomes.

Saporin has been used to construct fusion proteins with a variety of cytokines. These fusion proteins are bifunctional reagents that specifically bind to receptors by means of their growth factor moiety. The fusion protein/receptor complexes are then internalized by the cells, which are killed subsequently by the toxic moiety of the fusion protein (see also: Cytokine fusion toxins, Mitotoxins).
