Sodium orthovanadate (Na3VO4) at micromolar concentrations is a potent general inhibitor of phosphatases. Vanadate also inhibits phosphotyrosine phosphatases in cultured cells. Vanadate is not a complete inhibitor as some phosphatases are not inhibited. Most published reports show a residual (5-10 %) phosphotyrosine phosphatase activity in inhibitor concentrations of up to 1 mM vanadate.
Treatment of cells with this inhibitor results in an increase of proteins phosphorylated at tyrosine residues. Since transient phosphorylation of proteins on tyrosine is an important step in receptor mediated signal transduction of many cytokines (see also: PTK; protein tyrosine kinase) this agent can be used to mimic cytokine
